Abstract
Purpose: At present there is no alternative therapeutic way to diet and enzyme treatment for inherited metabolic diseases. Embryonic stem (ES) cells have strength to treat such genetic diseases since they differentiate into diverse tissues and proliferate in unlimited numbers without differentiation. This study aims to develop and test the feasibility of the new therapy using mouse model system by establishing the culture condition for the differentiation of ES cells in vitro and in vivo, the purification of differentiated ES cells, the method of transplantation, the establishment of self-ES cell by nuclear transfer, the gene manipulation of ES cells.
Methods : a. Establishment of culture condition for the proliferation and differentiation of ES cells b. Histological and biochemical study of the differentiated ES cells cultured in vitro and in vivo.
Results : The condition for the proliferation and differentiation of ES cells were established. Among methods we tested, the suspension culture was the best way for the differentiation of ES cells based on the expression of albumin (ALB) transcript. The ES cells were differentiated in suspension for 5 weeks and at the time interval of 1 week the differentited ES cells were analyzed for the expression of ¥á-fetoprotein (AFP) and ALB The expression of ALB and AFP increase gradually during the suspension culture. The differentiated ES cells were implanted into the spleen of nude mouse and in the ES cell-derived teratoma hepatocyte-like cells were detected and confirmed by the hepatocyte-specific antibodies.
